چکیده
Ramie (Boehmeria nivea L. Gaudich) is a major natural fiber crop cultivated in East Asia. The improvement of its yield and fineness are important breeding goals. Fiber complex quantitative trait comprising ramet number, stem diameter, plant height, skin thickness, percentage. crucial for ramie quality. However, there few association studies traits on ramie, lack high-density SNP maps population. Here, panel 112 core germplasms were genotyped by 215,376 consistent single-nucleotide polymorphisms (SNPs) from specific-locus amplified-fragment sequencing (SLAF-seq), used genome-wide study yield. Subsequently, the genetic diversity, linkage disequilibrium (LD), population structure was conducted based SNPs. Population cluster analysis disclosed five subpopulations. Neighbor-joining (NJ) revealed three clusters. No obvious relationships identified between them their geographic origins. LD decayed to r2 = 0.1 ~ 11.75 kbp physical distance. One, seven, one, twenty-seven significant marker associations detected (third season), diameter thickness percentage (second respectively. Two promising candidate genes, whole_GLEAN_10029622 whole_GLEAN_10029638 resided trait-SNP which annotated as cotton fiber-expressed protein an Arabidopsis thaliana homebox ATH1, respectively validated qPCR. loci or genes may provide basis future research marker-assisted selection ramie.

Genome-wide association study of rust traits in orchardgrass using SLAF-seq technology
BACKGROUND While orchardgrass (Dactylis glomerata L.) is a well-known perennial forage species, rust diseases cause serious reductions in the yield and quality of orchardgrass; however, genetic mechanisms of rust resistance are not well understood in orchardgrass. RESULTS In this study, a genome-wide association study (GWAS) was performed using specific-locus amplified fragment sequencing (SL...
متن کاملTranscriptome Analysis of Ramie (Boehmeria nivea L. Gaud.) in Response to Ramie Moth (Cocytodes coerulea Guenée) Infestation
The ramie moth Cocytodes coerulea Guenée (RM) is an economically important pest that seriously impairs the yield of ramie, an important natural fiber crop. The molecular mechanisms that underlie the ramie-pest interactions are unclear up to date. Therefore, a transcriptome profiling analysis would aid in understanding the ramie defense mechanisms against RM. In this study, we first constructed ...
متن کاملMolecular Cloning, Recombinant Expression and Antifungal Activity of BnCPI, a Cystatin in Ramie (Boehmeria nivea L.)
Phytocystatins play multiple roles in plant growth, development and resistance to pests and other environmental stresses. A ramie (Boehmeria nivea L.) phytocystatin gene, designated as BnCPI, was isolated from a ramie cDNA library and its full-length cDNA was obtained by rapid amplification of cDNA ends (RACE). The full-length cDNA sequence (691 bp) consisted of a 303 bp open reading frame (ORF...
متن کاملDevelopment and validation of molecular markers for characterization of Boehmeria nivea var. nivea and Boehmeria nivea var. tenacissima
BACKGROUND The root of Boehmeria spp (ramie) is a hepatoprotective Chinese herbal medicine. Medicinal properties vary between Boehmeria nivea var. nivea and Boehmeria nivea var. tenacissima, which are local species found in Taiwan. As commercial preparations may use either species, there is a need for a rapid and simple assay to identify variants for quality control. METHODS Four methods were...
متن کاملTranscriptome Sequencing and Profiling of Expressed Genes in Phloem and Xylem of Ramie (Boehmeria nivea L. Gaud)
Ramie (Boehmeria nivea L. Gaud) is a highly versatile herbaceous plant which is widely cropped in southern China. The success of this herbaceous plant relies on wide use in modern industry. Understanding the profiling of expressed genes in phloem and xylem of ramie is crucial for improving its industrial performance. Herein, we uncover the transcriptome profile in phloem and xylem in present st...
